A college founded in 1479 by Cardinal Pedro González de Mendoza in accordance with the papal bull issued by Sixtus IV.

Work started in the Gothic style but the Cardinal, enthused by the Renaissance, did not take a liking to it and changed the architect and work continued in the new Renaissance style. In 1761 Ventura Rodríguez made designs to substitute the large Gothic windows on the main facade for contemporary neoclassical ones. This was carried out by his pupil Manuel Godoy, who also transformed the openings of the three facades, finishing in 1748.

Nowadays the building houses the Rectoral organs of the university and the Historical Library that contains the Colegio del Cardenal library's bequest. The guest quarters are the actual Colegio Mayor de Santa Cruz (university residence). It was built in 1675 by Francisco de la Torre and has a square courtyard in two parts on pilasters and round arches cut in stone.
