El punto de partida de la Ruta de los Místicos es la ciudad de Valladolid, donde Santa Teresa fundó el cuarto de sus conventos de carmelitas descalzas, en cuya iglesia se guardan esculturas de Gregorio Fernández y obras de arte excepcionales en el claustro. Además conserva la celda que ocupara la Santa en sus estancias y el autógrafo original de su Camino de Perfección.
